In this GE Molecular Discovery role you will focus on developing new tools for our core platforms. With your help, we will expand and enable our platforms for non-viral gene therapy applications for single modality cures in a range of genetic diseases.
Position Summary
Poseida is seeking an outstanding candidate to join our Genetic Engineering team. The GE Molecular Discovery Scientist will play a key role focusing on expanded use, capabilities, and evolution of our core RNA-guided nuclease (Cas-CLOVER) and transposon (piggyBac) platforms.
Responsibilities
These may include but are not limited to:

• Expand our proprietary gene editing platform to develop new tools and/or applications for specialized and enhanced implementation in gene therapy and gene editing therapeutics.
• Apply your strong background in molecular biology, cancer biology, vectorology, or RNA-guided nucleases (RGNs) to develop new gene editing tools in cell culture models for translation to in vivo therapies.
• Assist with development and advancement of nascent and active pipeline for Poseida gene therapy products.
• Work in a highly collaborative, cross-functional team environment to perform experiments in the lab, analyze results, and communicate findings in team meetings.
• Keep current on trends, new products and technologies in gene editing and gene therapy.
• May manage 1-2 Research Technicians or Research Associates.

Requirements: Requirements, Knowledge, Skills and Abilities

• PhD in molecular biology, cell biology, cancer biology, virology or related field with 0 to 5 plus years of hands-on experience with RGNs, ZFNs, TALENs, DNA transposons, or vectorology (an equivalent combination of education and experience may be considered).
• Extensive expertise with advanced molecular biology and cloning methodology is required.
• Prior experience with in vitro non-viral gene delivery (transposon, episomal, or nanoparticle) is strongly preferred, especially regarding experience and/or expertise with DNA transposon technology.
• Expertise in the culture and genetic manipulation of primary cells, including hepatocytes, HSCs, hematopoietic cells (lymphocytes/monocytes), hPSCs or other cell types is a plus.
• Previous experience with functional assays used for measuring success of gene editing/knock-in/knock-outs/transgenesis (e.g. via live cell reporter assays, Western blot, cellular phenotypes, or next-generation sequencing).
• Previous experience with mammalian expression vector design, promoter screening/profiling, genetic logic gates, synthetic biological switches, or other conditional expression modules is a plus.
• Intimate knowledge or direct experience using directed evolution strategies, FGS, or variant library screens in either eukaryotic or prokaryotic contexts is a plus, although not required.
• Proficiency with multi-parameter flow cytometry, cell sorting, and FACS analysis software is a plus.
• Prior experience with plasmid (viral or non-viral) library preparation and amplification is a plus.
• Must possess excellent oral and written communication skills
